Concentration = vial strength divided by the bacteriostatic water added. On a U-100 syringe 100 units = 1 mL, so units = (dose divided by concentration) x 100. Values are syringe units, not milligrams. Amber values exceed 100 units, which is more than one standard 1 mL U-100 syringe holds, so that combination needs a more concentrated solution. Frequently asked questions
Wegovy, Ozempic, and Rybelsus are FDA-approved — so why calculate a semaglutide dose by hand?
Because this tool isn’t for any of those products. The branded injectables (Wegovy and Ozempic, cleared under FDA label 218316) ship as prefilled pens that are already dose-metered, and Rybelsus is an oral tablet on a different dose scale that is never drawn into a syringe. Hand math only comes up with self-reconstituted lyophilized vials — the compounded or gray-market kind that arrive as dry powder with no dose markings. This page converts a target mg dose into U-100 syringe units for those vials, and nothing else.
How do 5 mg, 10 mg, and 30 mg semaglutide vials relate to the 0.25–2.4 mg dosing range?
Those milligram figures describe the total drug in the vial, not one dose. The FDA label describes weekly injectable doses running from 0.25 mg up to 2.4 mg, so a 5 mg vial holds several weeks’ worth even at the top of that range, and many more at the 0.25 mg starting amount. The calculator divides whichever vial strength you enter (5, 10, or 30 mg) across your reconstitution volume to tell you how many U-100 units equal your target dose.
